Today there is a great interest in developing technologies to
precisely organise and integrate proteins into highly ordered micro-
and nanostructured assemblies. One approach to achieve this goal is
the use of DNA directed immobilisation ( DDI) which makes use of the
specificity of DNA base pairing to anchor protein to surfaces of
planar substrates or nanoparticles and offers many advantages over
conventional immobilization methods. Covalent heme-DNA adducts can be
used to prepare semisynthetic DNA- heme enyzme conjugates containing
DNA oligomers coupled in close proximity to the active site. Such
conjugates show remarkable kinetic activity and are capable of
specific binding to a solid supports containing capture DNA probes.
The programmable binding properties enabled by DNA hybridisation make
such semisynthetic enyzmes useful for a broad range of applications,
particulary in biocatalysis, electrochemical sensing and as building
blocks in biomaterial design. In addition heme enyzmes, which play a
key role in numerous applications in the field of biocatalysis and
bioanalyitics can be coupled to quantum dots (QD) nanoparticles and
their activity reversibly switched on an off by photoirradiation.
Four different peroxidases were successfully activated using this QD-
irradiation methodology which is anticipated to prove very useful in
biosensing, biocatalysis and design of novel cellular assay procedures.
